Do you think a man who's getting tickled mercilessly could break out of leather cuffs if he was strong enough?

It is highly unlikely. Unless, of course, the shackles were old or weak. Maybe the buckles were weak or some such otherwise. But, if the shackles were of good quality, new, and the buckles were solid, you're more likely to yank the rope or otherwise off if they're not tied properly or the rope was weak. In any case, as long as everything is solid and new and/or well tied, I personally don't see it happening.

I suppose it's possible but I dont think it's very likely.

Yes. I've managed to wriggle so much that the buckles came undone.
Rope tends to be a little better (if a bit less comfortable), locking cuffs are probably the best.

I once broke a cheap leather wrist cuff at the Hidden Chamber when Mistress Zara worked there. The rivets came right out. But nobody could break out of the leather cuffs I've got now. (They resemble the ones shown below but are a different color.)
